In principle, the PCR laboratory consists of four separate work areas—reagent preparation, sample preparation, amplification, and amplification product analysis—served by a dedicated corridor. Buffer rooms should be established for each work area, and interlocking mechanisms should ideally be installed between work areas and their respective buffer rooms. Functionally distinct work areas must be physically separated and clearly marked; there should be no direct access between them, and if they are closely adjacent, pass-through boxes must be installed. The first two areas constitute the pre-amplification zone, while the latter two constitute the post-amplification zone; these zones must be strictly separated. Materials, reagents, records, pens, cleaning supplies, and the like must flow only from the pre-amplification zone to the post-amplification zone (i.e., reagent preparation → sample preparation → amplification → product analysis) with no reverse flow.
Unidirectional flow—covering experimental processes, materials, personnel, and air—must be maintained between areas to create a protective barrier, prevent cross-interference, and avoid contamination caused by aerosol dispersion. The laboratory enclosure structure must be robust and airtight; all internal corners (both concave and convex) should feature coved (radiused) transitions. Interior wall surfaces must be smooth, dust-resistant, corrosion-resistant, and easy to clean and disinfect. Flooring should consist of PVC sheet flooring or self-leveling epoxy resin, meeting requirements for seamlessness, leak-proof integrity, smooth finish, and corrosion resistance.
