Cell culture is an aseptic technique requiring a working environment free from microorganisms and external interference. To prevent microbial contamination, the cell culture room must be clean, with air that is fresh, dry, and dust-free.
The cell culture laboratory comprises six sections: the aseptic operation area, incubation area, preparation area, storage area, and cleaning/sterilization area. These areas share common characteristics: spaciousness and brightness; easy-to-clean, non-slip flooring; smooth, hard wall surfaces; a logical equipment layout that facilitates operation and prevents cross-contamination; and a minimalist design that simplifies cleaning.
An ideal aseptic operation suite should be divided into three sections:
a) Changing room: For changing clothes and shoes, and putting on caps and masks.
b) Buffer room: Located between the changing room and the operation room; used for preparatory work and contamination prevention to maintain the sterility of the operation room. It can also house constant-temperature incubators and essential small instruments.
Aseptic operation room: Dedicated to aseptic procedures and cell culture. It should be appropriately sized, with a ceiling height that is not excessive, ensuring the effectiveness of UV sterilization. Walls must be smooth to facilitate cleaning and disinfection. As the room is small and sealed, air disinfection is typically achieved via UV lamps, though some laboratories employ ozone-free UV lamps or electronic sterilizers.
